Because the chelate is stable, it is gentle on the stomach and is partly absorbed through the gut’s amino-acid pathways.7 In a 60-day trial, an amino-acid chelate of magnesium raised blood magnesium where magnesium oxide did no better than placebo,8 and oxide was only about 4% absorbed in another study.9 The European Food Safety Authority rated magnesium bisglycinate not of safety concern.10 The 440 mg serving equals India’s full daily magnesium allowance for adult men.11

What buffering quietly takes away

Pure chelate means the full glycine dose.

Glycine only comes with magnesium that is properly chelated. When a product is “buffered” with magnesium oxide, part of its magnesium arrives with no glycine at all.

Take a typical buffered tablet listing 2,000 mg of “magnesium glycinate” and 440 mg of magnesium. To reach that number, roughly half its magnesium has to come from oxide — leaving only about 1.4 g of glycine. How to spot unbuffered magnesium glycinate on any label.

  • Do the math. Divide the magnesium by the glycinate weight. About 12–14% means a true chelate. 18% or more means oxide has been added.
  • The word “buffered”. Also watch for “chelate blend” or “glycinate complex”. Look for “fully reacted”, “non-buffered” or “unbuffered”.
  • “Magnesium oxide” in the ingredients. It’s cheap and was only about 4% absorbed in one study.9 In a glycinate product, it means buffering.
  • The word “elemental”. “2000 mg magnesium glycinate” is compound weight. Check how many milligrams of actual magnesium you get.
  • Many forms, no amounts. “Triple” or “multi-form” magnesium without a per-form amount hides how much is oxide.
  • “Proprietary blend”. If the doses are hidden, you can’t tell what you’re taking.

What is non-buffered magnesium glycinate?

It is magnesium glycinate with nothing mixed in. Every magnesium atom is bound to two glycine molecules (a “fully reacted” chelate). Many products are buffered — blended with cheaper magnesium oxide so the label shows more magnesium per tablet. Non-buffered means all the magnesium comes from the gentle, well-absorbed glycinate form.

How can I tell if a magnesium glycinate is non-buffered?

Yes. “Bisglycinate” is the more exact chemical name — “bis” means two glycine molecules per magnesium atom. Magnesium glycinate and magnesium diglycinate refer to the same compound.

How much glycine does it contain, and why does it matter?

About 2.7 g of glycine per three-tablet serving, carried by the magnesium chelate itself. Glycine is an amino acid your brain uses as a calming signal, and human studies used 3 g before bed for sleep quality. A buffered product at the same magnesium dose carries roughly half as much glycine, because the oxide part has none.

Why three tablets a day?

Because pure chelate is bulky. Getting 440 mg of magnesium takes 3,667 mg of magnesium bisglycinate, too much for one or two tablets. Products that fit 440 mg into two small tablets usually do it by adding oxide. Three tablets is the honest sign of a non-buffered formula.

Is 440 mg of magnesium a day safe?

Yes, for healthy adults. 440 mg is India’s full daily magnesium allowance for adult men (ICMR-NIN 2020). The women’s allowance is 370 mg, so women can take two tablets (about 293 mg). Higher magnesium doses can loosen stools; if that happens, drop a tablet. If you have kidney disease, are pregnant or breastfeeding, or take prescription medication, check with your doctor first.

When is the best time to take it?

Mostly in the evening. One tablet with dinner and two about an hour before bed works well, or take all three in the evening. Take them with food if your stomach is sensitive.

Will it help me sleep?

It supports your evening wind-down. Magnesium bisglycinate is the form used in a 2025 trial where adults with poor sleep improved their insomnia scores more than on placebo within four weeks, and glycine has been studied before bed for sleep quality. It is not a sleeping pill, and results vary — good sleep habits still matter most.

Will it upset my stomach?

It’s designed not to. There is no magnesium oxide, the form most linked to loose stools, and magnesium bisglycinate was better tolerated than oxide in a clinical study. If you’re sensitive, take it after food.

Can I take it with other supplements or medicines?

Yes, with a few rules. Space it at least two hours from tetracycline or quinolone antibiotics, bisphosphonates and thyroid medication. Add up the magnesium from all your supplements so your total stays sensible. If you take blood-pressure or other prescription medicines, ask your doctor first.

Is it vegetarian? What about allergens?

Yes, the tablets are vegetarian. The ingredients contain no gelatin, sugar or proprietary blends. They are made in a facility that also processes oats, milk, soy, wheat and nuts.

How soon will I notice a difference?

It depends on where you start. Magnesium trials have typically measured changes over four to eight weeks of daily use, and people who eat less magnesium tend to notice more. Consistency matters more than anything else.

About the studies
  • The 2025 sleep trial gave 250 mg of magnesium a day as bisglycinate to 155 healthy adults with poor sleep; this product gives 440 mg. The improvement was modest, and larger in people who ate less magnesium.1
  • The glycine sleep studies gave 3 g of pure glycine before bed. They were small (11 people in the sleep-lab study) and funded by a glycine maker.2,3 The body-cooling pathway was mapped in rats.12 Here the glycine is carried by the magnesium chelate rather than taken on its own.
  • The stress trial used a different magnesium salt in adults with low blood magnesium.5 The muscle-soreness trial was in 22 young adults over 10 days, at 350 mg a day.6
  • The stomach-tolerance study was in 12 people after bowel surgery: overall absorption was similar to oxide, and the chelate absorbed better in the four people who absorbed oxide worst.7 The 60-day comparison used an amino-acid chelate of magnesium, not specifically bisglycinate.8
  • The “buffered” glycine example assumes pure bisglycinate is about 14% magnesium and magnesium oxide about 60%; real buffered products vary.
  • For supplements, Europe sets 250 mg and the US 350 mg of magnesium a day as levels to avoid loose stools in everyone. India’s ICMR-NIN allowance is 440 mg for men and 370 mg for women, from all sources.11
